Cytotoxic, immunosuppressive and trypanocidal activities of agrocybin, a polyacetylene produced by Agrocybe perfecta (Basidiomycota)

摘要: The ethyl acetate extract from the culture of fungus Agrocybe perfecta (Rick) Singer was selected for further study in a screening Brazilian basidiomycetes bioactivity. showed significant activity against recombinant enzyme trypanothione reductase (TryR) Trypanosoma cruzi, lymphocyte proliferation human peripheral mononuclear cells (PBMC) stimulated with phytohemaglutinin (PHA), and cancer cell lines UACC-62 (melanoma), MCF-7 (mammary), TK-10 (kidney). chromatographic fractionation monitored by above bioassays that agrocybin active component. Agrocybin, known polyacetylene amide, an IC50 2 μM TryR assay but killed only 60% trypomastigote form T. cruzi infected murine blood even at 680 μM. This weaker could be due to low temperature used mimic banked or as consequence its inactivation blood, already reported literature. On other hand, it inhibited PBMC 50% 3.4 μM growth concentrations between 9 24.5 μM. Measurements DNA fragmentation using flow cytometry suggest promotes death via apoptosis.
