Effects of catholytes on the mobilization of proteins after capillary isoelectric focusing

摘要: Purified proteins and human plasma were separated by capillary isoelectric focusing detected cathodic mobilization, e.g. replacing the sodium hydroxide solution solutions containing another anion. The effect of catholyte anions on resolution mobilized was examined. Compared with chloride or phosphate anion, organic having small dissociation constants improved protein resolution, especially in range acidic proteins. Among catholytes examined, acetic acid gave best for purified Measurement changes electric current during mobilization process indicated that low mobility move slowly towards anode retard pH gradient capillary. Isoelectric focusing, direct measurement polyacrylamide gel columns (1.3 mm internal diameter 38 long) also revealed these effects anions.
