Repeated KI Prophylaxis in Case of Prolonged Exposure to Iodine Radioisotopes: Pharmacokinetic Studies in Adult Rats

摘要: To propose a new and effective dose regimen for stable potassium iodide (KI) repeated prophylaxis in case of prolonged exposure to radioactive iodine. The pharmacokinetics iodine was determined rats by compartmental analyses after intravenous oral administrations the optimal 1 mg/kg KI, which previously selected dose-effect study. thyroid protection against iodine-125 incorporation followed during 24 h single dosing KI. A KI modeled using initial estimates pharmacokinetic parameters. consisting daily 8 days studied. Plasma concentrations predicted simulation were verified experimental data varied third between 174 1190 μg/l. inhibition study binding as function time showed that effect could be correlated plasma concentrations. Hence, theoretical decrease uptake from 63 88% achieved 24 h-interval two doses. Given satisfactory levels protection, this envisaged order extent indications prophylaxis.
