摘要: Exposure to arsenic in its inorganic form, arsenite, causes adverse effects many different organs and tissues. Here, we have investigated arsenite-induced on vascular tissues the model organism zebrafish, Danio rerio. Zebrafish embryos were exposed arsenite at exposure windows susceptibility tissue damage was recorded 72hours post fertilization (hpf). Intersegmental vessel sprouting growth most perturbed by during 24-48hpf window, while disruption condensation of caudal vein plexus more often observed 48-72hpf reflecting when these structures develop normal embryogenesis. The rate decreased exposure, deviated from that control around 24-26.5hpf. We further mapped changes expression key regulators angiogenesis vasculogenesis. Downregulation endothelial factor receptor 1/fms-related tyrosine kinase 1 (vegfr1/flt1) evident already 24hpf, coinciding with rate. At later time points, matrix metalloproteinase 9 (mmp9) upregulated, suggesting affects composition extracellular matrix. In total, eight factors involved aspects vascularization significantly altered exposure. conclusion, our results show is a potent disruptor developing zebrafish embryo, finding calls for an evaluation as developmental toxicant mammalian systems.
