Low pH value of pericellular medium as a factor limiting cell proliferation in dense cultures

摘要: An attempt was made to determine the role of metabolic acidification pericellular medium in regulating cell proliferation. A method measuring pH at a distance exceeding Debye radius 5–10 from surface (pHp) developed. The values pHp and measured >1 cm cells (pHm) were found differ, depending on population density. At density least 7 × 105 cells/cm2 (maximum saturation density) pHm 7.4–7.6, reached value ~6.5. It that 6.5 unfavorable for proliferation sparse cultures, where equal. Based these findings, low as revealed present work using dense cultures optimal can be considered limiting factor multiplication.
