Toxin-produced Purkinje cell death: A model for neural stem cell transplantation studies

摘要: Purkinje cell loss is the hallmark of cerebellar ataxias. Here fungal neurotoxin Penitrem A was used to create partially Purkinje-cell-deficient cerebella in neonate and young adult rats suitable for use neural stem transplantation studies. I.p. administration P3, P6 11-week old caused noticeable tremor all treated animals that lasted between 1 3 days more immediate rat pups than rats. Quantification cresyl violet stained sections showed cells were preferentially lost vermis specifically folia VI IX (P<0.001-0.05). No change occurred number I-III folium X. These results confirmed by calbindin binding layer appearance enlarged vacuolated mitochondria. The present study show can remove immature cerebellum thus provide a potential model micro-environmental cues vivo differentiation from transplanted and/or intrinsic cells.
