Isolation of the C-type lectin like-domain cDNAs from the mud crab Scylla paramamosain Estampador, 1949, and its expression profiles in various tissues, during larval development, and under Vibrio challenge

摘要: Scylla paramamosain Estampador, 1949 is one of the most precious marine crabs farmed in China. The crab prone to infection by microbes at various stages during its growth, leading high mortality. C-type lectin has a characteristic carbohydrate recognition domain and plays an important role immunity system. A cDNA library from mud S. was constructed using SMART technique. Two complete sequences showing identities with gene were isolated library. two full-length cDNAs (SpLec1 SpLec2) consist 746 834 bp, respectively. Quantitative realtime PCR analyses revealed that lectins expressed mainly haemocytes, muscle hepatopancreas, highest expression level found hepatopancreas. mRNA continuous developmental zoeal phases also examined quantitative real-time PCR. After Vibrio parahaemolyticus (Sakazaki, 1963) concentration 2.3 × 10 6 cfu/ml, temporal SpLec1 SpLec2 megalopa stage first increased, reached maximum, then dropped original again. research could shed new light on studies moulting crab.
