摘要: Abstract IL-16 is a pro-inflammatory cytokine originally designated as lymphocyte chemoattractant factor. In mammal and avian, it has been characterized an essential regulator of various cellular processes including cell recruitment activation against pathogen invasion. So far, neither the full-length homologue nor response mechanism was reported in crab species. present study, pro-IL-16 firstly cloned from mud Scylla paramamosain . The Sp-pro-IL-16 consisted 4107 bp with opening reading frame encoding 1369 amino acids. Multiple alignment analysis showed putative acid sequence had about 73.86% identity Litopenaeus vannamei pro-IL-16. Additionally, two conserved PDZ domains protein binding sites were found Sp- high similarities 94.19% 51.14% their Mus musculus counterparts. RT-PCR indicated that transcripts constitutively expressed all tissues examined extreme level hepatopancreas. Moreover, hepatopancreas significantly up-regulated 15-fold at 72 h after Vibrio alginolyticus challenge 3.5-fold 12 h virus-analog Poly (I:C) challenge. Western blot revealed can be cleaved to its bioactive form, approximately 35 kDa mature IL-16, levels both increased It first experimental identification arthropods. This study could shed new light on further understanding pathogens. Meanwhile, brought insight into origin evolution
