摘要: NFIL3 is a transcriptional activator of the IL-3 promoter in T cells. In vertebrates, it has been characterized as an essential regulator several cellular processes such immunity response, apoptosis and NK cells maturation. However, identification functional characterization still remains unclear arthropods. this study, homologue was firstly cloned mud crab Scylla paramamosain. The full-length SpNFIL3 2, 041 bp length with open reading frame 1, 509 bp, containing conserved basic region leucin zipper domain. qRT-PCR analysis indicated that significantly highly expressed hepatopancreas hemocytes. Moreover, transcription could be up-regulated after challenge Vibrio alginolyticus or virus-analog Poly (I:C). dual-luciferase reporter assays revealed activate NF-κB pathway. immunofluorescence assay located nucleus. After interfered vivo vitro, expressions two members (SpRelish SpDorsal), six antimicrobial peptide genes (SpCrustin SpALF2-6) pro-inflammatory cytokine SpIL-16 were suppressed, bacteria clearance capacity crabs also markedly impaired silenced crabs. These results played crucial role innate S. paramamosain brought new insight into origin evolution arthropods even invertebrates.
