摘要: This study was aimed at exploring the effects of P2X7 receptor on BV2 microglia cell injury induced by glycoprotein gp120 (gp120) and its underlying mechanisms. We used MTS method to influence different concentrations cells, test degree in each treatment group; quantitative real-time PCR (qPCR) Western blot were detect mRNA protein expressions. Immunocytochemistry expression P65 NF-κB, respectively. also measured content TNFα, IL-1β, nitric oxide (NO) reactive oxygen species (ROS). found that survival rate generally decreased as concentration increased, + Brilliant Blue G (BBG) group higher than group. qPCR results showed expressions positively dose-dependent with concentration; immunocytochemistry NF-κB increased significantly compared those control (Ctrl) group, but gp120+BBG decreased. Taken together, these confirmed is involved gp120-induced microglial mechanism may be associated over-activation caused up-regulation, which leads abundant release inflammatory factors exert toxic cells.
