The HLA-A mutation assay: improved technique and normal results.

摘要: The HLA-A assay for detection of mutant lymphocytes has been modified, principally by incorporation an overnight-incubation step which resulted in improved immunoselection. For 83 estimations on cells from 21 normal individuals with a mean age 35, the vivo frequency was 2.99 X 10(-5) +/- 1.48 (mean 1 SD). 11 10 elderly 78, significantly greater, being 7.16 4.06 10(-5). Similar results were obtained when HLA-A2 or HLA-A3 mutants enumerated. Mutant measured at locus is almost order magnitude greater than that hypoxanthine phosphoribosyl transferase locus, but least major part this difference likely to be due factors other genetic stability between two loci.
