摘要: Cyclin D1 is a cell cycle regulator essential for G1 phase progression and candidate proto-oncogene whose deregulated expression has been implicated in pathogenesis of several types cancer. We have examined cyclin 212 primary tumours five histogenetically distinct by immunohistochemistry found strong aberrant accumulation the protein 21%, moderate overabundance further 25% cases. While abnormalities were more frequent carcinomas breast, i.e. cancer type known gene amplification, was also seen significant subsets colorectal cancers, soft tissue sarcomas, uterine malignant melanomas. Comparison stages tumour showed concordant patterns situ vs invasive breast carcinoma components (n = 37) between metastatic lesions 51) types. The specificity immunohistochemical data supported immunoblotting analysis lysates, tumour-specific over-expression confirmed computer-assisted image analysis. These observations suggest that alterations represent common feature malignancies diverse histogenesis indicate both spectrum frequency aberrations significantly exceed previous estimations based on genetic analyses.
