Autoradiographic studies on the uptake of 3 H‐dopamine by neurons and astrocytes in explant and primary cultures of rat CNS: effects of uptake inhibitors

摘要: The cellular localization of the uptake 3H-dopamine was studies in explant and primary cultures from various regions rat central nervous system by means autoradiography. In substantia nigra, taken up cell bodies processes many neurons. striatum, cerebellum spinal cord, neuronal were not labelled, whereas outgrowing nerve fibres revealed intense monoamine. Uptake neurons Na(+)- temperature-dependent, suggesting an active mechanism. cultures, astrocytes did accumulate 3H-dopamine, which prepared same as also this intensity labelling dependent on incubation time. Little observed after time 5 min only 10-15 show moderate labelling. indicating that glial cells do possess mechanism for This is consistent with biochemical investigations other laboratories, demonstrating a facilitated diffusion system. Addition inhibitors nomifensine or GBR 12909 to markedly reduced inhibited at concentration 10(-6) M. contrast, accumulation slightly affected At highest used (10(-5) M), blocked astrocytes.
