7β-Hydroxycholesterol Blocked at C-3-OH Inhibits Growth of Rat Glioblastoma In Vivo: Comparison between 7β-Hydroxycholesteryl-3β(ester)-oleate and 7β-Hydroxycholesteryl-3β-O(ether)-oleyl

摘要: Background: It was previously demonstrated that the 7‚-hydroxycholesteryl-3‚(ester)-oleate (7‚-ester) possesses antitumor properties against experimental rat C6 glioblastoma. The effect of an analog this molecule, 7‚-hydroxycholesterol-3‚-O(ether)-oleyl (7‚-ether), investigated. Materials and Methods: Liposomes containing no oxysterol (control), 7‚-ether or 7‚-ester were injected into tumors induced by cells in brain cortex. At defined times, animals sacrificed, stained with cresyl violet their volumes measured densitometry. Oxysterol clearance assessed quantification from lipid extraction treated tumors. Results: new compound slower than form. forms displayed similar activities 3-day-old In contrast, form more active on well-developed glioblastoma: 75 nmol inhibited tumor growth 70% compared to controls, while had under such conditions. a cytostatic rather cytotoxic effect. addition, composition liposomes did not affect activity. Conclusion: Only blockade C-3-OH group is required for kind oxysterol.
