Quantitation of purified monoclonal antibody needed to prevent HSV-1 induced stromal keratitis in mice.

摘要: A purified lgG2a monoclonal antibody with a neutralizing titer of 10 and specificity for gD was evaluated its therapeutic potential in murine ocular infection model. BALB/c mice, infected on the scarified cornea times HSV-1 strain RE concentration needed to produce severe persistent stromal opacity, were given single inoculation intraperitoneally 24 hours later. The animals then followed corneal disease development. Antibody, at concentrations as low μg per mouse, strikingly effective preventing opacity. Furthermore, corneas, once clear, remained clear whereas controls developed +3 +5 which still present 60 days post-infection. Animals that had been treated recovered from resistant subsequent challenge opposite cornea. These results demonstrate systemically administered microgram quantities anti-gD antibody.
