The SOS chromotest: Direct assay of the expression of gene sfiA as a measure of genotoxicity of chemicals

摘要: Resume Nous avons utilise une fusion genetique qui met l'expression du gene lacZ de la β-galactosidase sous le controle promoteur sfiA, pour construire nouvelle souche bacterienne. Celle-ci permet d'examiner, en quelques heures, capacite d'un produit a endommager DNA, grâce un simple dosage colorimetrique. Les courbes « dose-reponsecomportent region lineaire definir pouvoir inducteur (SOSIP) des produits. Sur serie produits examines, nous observe valeurs SOSIP s'etendent sur 7 decades et sont etroitement correlees avec mutagene evalue dans test d'Ames (Mutatest) entre d'induction prophage λ (Inductest). La sensibilite (plus faibles quantites detectees) est comparable SOS Chromotest Mutatest mais generalement plus faible l'Inductest. Nos resultats suggerent qu'au moins partie reponse observee depend l'induction d'une fonction que plupart genotoxiques inducteurs systeme SOS, c'est-a-dire qu'ils peuvent conduire l'activation protease RecA.
