Bioluminescent assays using glucose-6-phosphate dehydrogenase: application to biotin and streptavidin detection.

摘要: Abstract A streptavidin-glucose-6-phosphate dehydrogenase (G6PDH) conjugate was synthesized and its properties were studied, along with those of biotin-G6PDH conjugates. Two bioluminescent assays used. Streptavidin assayed in two steps: streptavidin samples first incubated a small amount then biotinylated rabbit γ-globulins. The complex immobilized on immunoadsorbent. In the single-step biotin assay, free allowed to compete linked γ-globulins for binding streptavidin-G6PDH presence Neither assay required washing or separation steps sensitivity 0.2 ng 100 fg biotin. Different applications are described: studies reactivity when probes solution immobilized, quantitation DNA oligonucleotides.
