摘要: The cDNA clone (pNP24) coding for a protein induced by exogenous NaCl has been isolated from tomato root library with the use of an inosine containing synthetic oligomer. authenticity established comparing sequence to NH2-terminal which purified homogeneity HPLC. nucleotide pNP24 reveals 5′ signal sequence, open reading frame 718 nucleotides, 3′ AT rich untranslated region probable polyadenylation and poly A stretch. mature polypeptide as deduced molecular weight 24226. This named NP24. It is slightly basic unusually high number cysteines (15). Northern blot analyses reveal that abundance mRNA NP24 at least 100-fold greater in suspension cells log phase grown medium than control medium. below level detection roots young plants until several weeks after germination but it earlier higher levels stressed 0.171 M NaCl. Thus salt stress accelerates accumulation message roots. comparison steady state immuno indicates this determined its level. not secreted localized within cytoplasm or soluble fraction nucleus, vacuole, microbodies. degree homology (58%) thaumatin, considerable value artificial sweetener.
