摘要: A new protocol is described for amplifying label-free electrochemical measurements of DNA hybridization based on the enhanced accumulation purine nucleobases in presence copper ions . Such electrical assays involve target to inosine-substituted oligonucleotide probes (captured magnetic beads), acidic dipurinization hybrid DNA, and adsorptive chronopotentiometric stripping free ions. Both amplified adenine guanine peaks can be used detecting hybridization. The dramatic signal amplification advantage this type detection has been combined with efficient removal non-complementary use microliter sample volumes disposable transducers. Factors influencing enhancement were assessed optimized. limit 40 fmol (250 pg) was obtained 10 min 5 adsorptive-accumulation times. advantages of procedure demonstrated by its application segments related BRCA1 breast cancer gene. holds great promise not only hybridization, but also trace measurement nucleic acids.
