Overexpression of Lin28b in Neural Stem Cells is Insufficient for Brain Tumor Formation, but Induces Pathological Lobulation of the Developing Cerebellum.

摘要: LIN28B is a homologue of the RNA-binding protein LIN28A and regulates gene expression during development carcinogenesis. It strongly upregulated in variety brain tumors, such as medulloblastoma, embryonal tumor with multilayered rosettes (ETMR), atypical teratoid/rhabdoid (AT/RT), or glioblastoma, but effect an vivo overexpression on developing central nervous system unknown. We generated transgenic mice that either overexpressed Lin28b Math1-positive cerebellar granule neuron precursors broad range Nestin-positive neural precursors. Sections vermis from adult Math1-Cre::lsl-Lin28b had additional subfissure lobule IV. Vermes p0 p7 Nestin-Cre::lsl-Lin28b appeared normal, we found pronounced vermal hypersublobulation at p15 p21 these mice. Also, external cell layer (EGL) was thicker than controls, contained more proliferating cells, persisted up to p21. Consistently, some Pax6- NeuN-positive cells were present EGL even p21, detected molecular (ML) compared control. Finally, residual Pax2-positive inhibitory interneurons ML which have already disappeared controls. conclude while does not lead formation, it results protracted leads vermis.
