摘要: Glycoproteins fulfill many indispensable biological functions, and changes in protein glycosylation have been observed various diseases. Improved analytical methods are needed to allow a complete characterization of this complex common post-translational modification. In study, we present workflow for the analysis microheterogeneity N-glycoproteins that couples hydrophilic interaction nanoreverse-phase C18 chromatography tandem QTOF mass spectrometric analysis. A glycan database search program, GlycoPeptideSearch, was developed match N-glycopeptide MS/MS spectra with glycopeptides comprised drawn from GlycomeDB structure peptide user-specified set potentially glycosylated peptides.
