摘要: In order to study the integrity of dialysis membranes pyrogens, dialysate side a closed loop hemodialysis (HD) circuit was challenged with E. coli microfiltrate containing 500 ng/ml endotoxin. Three solutions, a) tissue culture medium/saline, b) 5% human serum albumin, and c) 10% fresh plasma, were circulated in blood for five hours. Samples drawn from assayed interleukin-1 (IL-1)-inducing activity on mononuclear cells (MNC) vitro. No IL-1-inducing substances detected when saline or medium loop. Circulating albumin revealed samples only after hours HD. However, addition plasma resulted appearance an substance(s) 15 minutes After 30 minutes, maximum observed (control stimulation index, 3.30 +/- 0.67 SEM vs. 7.59 1.50, P less than 0.02). The completely inhibited by polymyxin B, cationic antibiotic which blocks Additional experiments demonstrated that vitro MNC IL-1-production induced same is enhanced presence plasma. These studies demonstrate that: (a) factors pass into compartment system bacterial pyrogen; (b) production IL-1 plasma.(ABSTRACT TRUNCATED AT 250 WORDS)
