Consolidated bioprocessing strategy for ethanol production from Jerusalem artichoke tubers by Kluyveromyces marxianus under high gravity conditions

摘要: Aims:  Developing an innovative process for ethanol fermentation from Jerusalem artichoke tubers under very high gravity (VHG) conditions. Methods and Results:  A consolidated bioprocessing (CBP) strategy that integrated inulinase production, saccharification of inulin contained in production sugars released by the enzyme was developed with inulinase-producing yeast Kluyveromyces marxianus Y179 fed-batch operation. The impact inoculum age, aeration, supplementation pectinase nutrients on performance CBP system studied. Although activities increased extension seed incubation time, its contribution to negligible because vigorously growing cells harvested earlier carried out more efficiently. Thus, overnight has been practised starch-based feedstocks is recommended. Aeration facilitated process, but compromised yield negative Crabtree effect species, increases risk contamination industrial conditions. Therefore, nonaeration conditions are preferred system. Pectinase reduced viscosity broth improved performance, particularly conditions, cost should be carefully balanced. Medium optimization performed, concentration as 94·2 g l−1 achieved when 0·15 g l−1 K2HPO4 supplemented, which presents a significant progress tubers. Conclusions:  using K. marxianus suitable efficient VHG conditions. Significance Impact Study:  alternative grain-based production. not only saves energy consumption distillation, also significantly reduces amount waste distillage discharged distillation
