The influence of one-carbon metabolism on gene promoter methylation in a population-based breast cancer study

摘要: Abnormal methylation in gene promoters is a hallmark of the cancer genome; however, factors that may influence promoter have not been well elucidated. As one-carbon metabolism pathway provides universal methyl donor for reactions, perturbation this might DNA and, ultimately, affect functions. Utilizing approximately 800 breast tumor tissues from large population-based study, we investigated relationships between dietary and genetic involved panel 13 cancer-related genes. We found CCND2, HIN1 CHD1 were most “dietary sensitive” genes, as their was associated with intakes at least two out eight examined. On other hand, some micronutrients (i.e., B2 B6) more “epigenetically active” intake levels correlated status 3 genes evaluated. Both positive (hypermethylation) inverse (hypomethylation) associations high micronutrient observed. Unlike what saw factors, did observe any clear patterns polymorphisms Our results provide preliminary evidence capacity to epigenome. Given epigenetic alterations are thought occur early development potentially reversible, modifications offer promising venues intervention prevention.
