摘要: Despite its importance in reproductive biology and women's health, a detailed molecular-level understanding of the human endometrium is lacking. Indeed, no comprehensive studies have been undertaken to elucidate important protein expression differences between endometrial glandular epithelium surrounding stroma during proliferative midsecretory phases menstrual cycle. We utilized laser microdissection harvest epithelial cells stromal compartments from secretory premenopausal tissue performed global, quantitative mass spectrometry-based proteomics analysis. This analysis identified 1224 total proteins cells, among which 318 were differentially abundant (q < 0.05), 1005 compartments, 19 0.05). Several chosen for validation by immunohistochemistry an independent set uterine tissues, including carboxypeptidase M, tenascin C, neprilysin, ectonucleotide pyrophosphatase/phosphodiesterase family member 3 (ENPP3). ENPP3, was elevated phase, confirmed be midsecretory-phase baboon lavage samples also observed N-linked glycosylated form that not phase. study provides view into global proteomic alterations cycling endometrium. These remodeling provide basis numerous follow-up investigations on function these regulated their role pathologies.
