Capillary isoelectric focusing-electrospray mass spectrometry for protein analysis

摘要: On-line combination of capillary isoelectric focusing (CIEF) with electrospray mass spectrometry (ESMS) as a two-dimensional separation system is demonstrated. Mixtures model proteins including cytochrome c (horse heart), myoglobin and carbonic anhydrase II (bovine erythrocyte) are focused cathodically mobilized in polyacrylamide-coated capillary. At the end CIEF capillary, protein zones analyzed by coupled on-line to an interface coaxial sheath flow configuration. The effects carrier ampholyte concentration on ionization spectra presented discussed. In this study, effect permits analysis very dilute samples. A typical factor 50-100 times observed. detection limit for full-scan CIEF-ESMS range 10{sup -7} M, 2 orders magnitude over that possible normal zone electrophoresis ESMS. 35 refs., 5 figs., tabs.
