Robust, high-throughput solution structural analyses by small angle X-ray scattering (SAXS)

摘要: We present an efficient pipeline enabling high-throughput analysis of protein structure in solution with small angle X-ray scattering (SAXS). Our SAXS combines automated sample handling microliter volumes, temperature and anaerobic control, rapid data collection analysis, couples structural archiving. subjected 50 representative proteins, mostly from Pyrococcus furiosus, to this found that 30 were multimeric structures solution. allowed us distinguish aggregated unfolded define global parameters oligomeric states for most samples, identify shapes similar 25 unknown structures, determine envelopes 41 proteins. believe is technology may change the way genomics research done.
