摘要: Abstract A Rhizopus oryzae lipase (ROL) has been expressed in Pichia pastoris as a reporter gene using two different regulated promoters. Both phenotypes, Muts and Mut+, have used for the expression under control of alcohol oxidase 1 promoter (PAOX1). Moreover, new formaldehyde dehydrogenase (PFLD1) tested. PFLD1 allows design methanol-free culture strategies, being methylamine less volatile flammable inducer. The comparison batch fed-batch cultures promoters shown that (PAOX1) is somewhat more efficient than Mut+ systems, with 1.1- 1.4-fold higher specific productivity (U lipase/g biomass h), respectively. However, all three systems was very similar (2500 U lipase/l h). These results demonstrate heterologous proteins transcriptional promising alternative to well-known PAOX1.
