Analyzing neurite outgrowth from explants by fitting ellipses.

摘要: The establishment of appropriate connectivity in the developing nervous system depends on many molecular guidance cues. A key method for studying response nerve fibers to such cues is culture explants neural tissue three-dimensional collagen gels. However, most previous analyses neurite outgrowth patterns from these have been very simple, often measuring only one or two parameters. Here we introduce a more sophisticated characterizing explants, based fitting an ellipse pattern outgrowth. This provides 5 parameters describing outgrowth: x and y position center ellipse, elongation, area tilt. We then apply this large dataset dorsal root ganglion grown presence precisely controlled gradients growth factor. analysis reveals number new features data.
