Netrin-1 Promotes Thalamic Axon Growth and Is Required for Proper Development of the Thalamocortical Projection

摘要: The thalamocortical axon (TCA) projection originates in dorsal thalamus, conveys sensory input to the neocortex, and has a critical role cortical development. We show that secreted guidance molecule netrin-1 acts vitro as an attractant growth promoter for thalamic axons is required proper development of TCA vivo. As TCAs approach hypothalamus, they turn laterally into ventral telencephalon extend toward cortex through population netrin-1-expressing cells. DCC neogenin, receptors implicated mediating effects netrin-1, are expressed whereas unc5h2 unc5h3, repulsion, not. In vitro, biased source which can be abolished by netrin-1-blocking antibodies. Netrin-1 also enhances overall outgrowth from explants thalamus. internal capsule zone telencephalic attenuated, but not significantly, antibodies, suggesting it releases another activity addition netrin-1. Analyses -/- mice reveal disorganized, their pathway abnormally restricted, fewer reach cortex. These findings demonstrate promotes cooperates with other cues control pathfinding thalamus
