Monoclonal antibodies to NF-Y define its function in MHC class II and albumin gene transcription.

摘要: NF-Y is a sequence-specific DNA-binding protein which, as heterodimer, recognizes CCAAT motifs in variety of transcriptional promoters. We have generated panel monoclonal and affinity-purified polyclonal antibodies directed against various epitopes NF-Y. These reagents are highly specific for either the A or B subunits; we mapped recognized by to glutamine-rich activation domain NF-YA. The inhibit vitro transcription from promoters albumin gene Ea, class II major histocompatibility complex. data definitively demonstrate role regulating two tissue-specific genes whose expression patterns do not overlap. Interestingly, cannot formed pre-initiation complex, but block reinitiation subsequent rounds same templates.
