摘要: The S/G2-specific transcription of the human cdc25C gene is due to periodic occupation a repressor element ('cell cycle-dependent element'; CDE) located in region basal promoter. Protein binding major groove CDE G0 and G1 results phase-specific repression activated transcription. We now show that CDE-mediated also principle underlying cyclin A cdc2 genes. single point mutation within 10- 20-fold deregulation an almost complete loss cell cycle regulation all three In addition, cdc25C, genes share identical 5 bp homology region'; CHR) starting at position, six nucleotides 3' CDE. Strikingly, CHR each promoters produces same phenotype as CDE, i.e. dramatic G0. agreement with these results, vivo DMS footprinting showed groove, minor groove. This applies particular presence NF-Y Sp1 sites which, gene, have been shown be targets through CDE.(ABSTRACT TRUNCATED AT 250 WORDS)
