The DDK inbred strain as a model for the study of interactions between parental genomes and egg cytoplasm in mouse preimplantation development

摘要: The DDK strain of mice has unusual genetic properties. When females this are crossed to males other strains, they generally exhibit a very low fertility, whereas reciprocal crosses fully fertile as the intrastrain crosses. observed fertility results from early embryonic lethality, F1 embryos dying around late morula-early blastocyst stage. Nuclear transplantation experiments between hybrid eggs BALB/c and strains shown that failure (DDK female x male) develop is not due combination per se maternal (DDK) paternal (BALB/c) genomes but rather an incompatibility genomic contribution cytoplasm. This does occur pronucleus cytoplasm, suggesting involvement differential imprinting parental genomes. Introduction cytoplasts isolated 1- 8-cell into male or corresponding developmental stage demonstrate cytoplasm prevents formation normal blastocysts through specific interaction with component diploid nucleus. Genetic molecular studies underway try isolate gene(s) responsible for male)F1 embryos. These should help in our understanding nucleocytoplasmic interactions respective roles development.
