摘要: Publisher Summary This chapter deals with the recent developments concerning isolation and characterization of DNA polymerases II III Escherichia coli. Genetic physiological experiments designed to elucidate biological function these enzymes are also discussed. The explores properties first polymerase be isolated from E. coli, polymerase. a mutant coli defective in I activity (PolA 1 - ) provided basis for several investigations into nature enzymatic mechanism replication such cells. After PolA mutant, it was generally assumed, since no other had yet been discovered that either residual amounts or some undiscovered perhaps unusual requirements required replication. Initially, we described an enzyme cells catalyzed synthesis DNA. resistant antiserum capable inactivating I. It there indeed exists another purification this enzyme, II, lysed after toluene detergent treatment by more conventional means were reported.
