摘要: Ristocetin cofactor activity is the test of choice for diagnosing von Willebrand disease. There however no simple and precise quantitative method its rapid determination in clinical laboratory. We describe here a new approach measuring ristocetin using an ELISA plate reader. Paraformaldehyde-fixed platelets, diluted plasma samples were mixed incubated wells microtiter plate. Platelet agglutination was measured by light absorbance at 405 nm. Measurements platelet 1x to 64x dilution normal pooled used construct standard curve. Further characterization showed significant linear correlation between results determined aggregometer assay (p < 0.0001), that intra- inter-assay variations 6% 17%, respectively. Therefore, newly developed provides way quantitation activity.
