摘要: The blood-brain barrier (BBB) is a major obstacle in drug delivery for diseases of the brain, and today there no standardized route to surpass it. One technique locally transiently disrupt BBB, focused ultrasound combination with gas-filled microbubbles. However, microbubbles used are typically developed imaging, not BBB disruption. Here we describe efficient opening using promising novel Acoustic Cluster Therapy (ACT), that recently has been Abraxane® successfully treat subcutaneous tumors human prostate adenocarcinoma mice. ACT based on conjugation liquid oil microdroplets through electrostatic interactions. Upon activation an field, microdroplet phase transfers form larger bubble lodges microvasculature. Further insonation induces volume oscillations activated bubble, which turn induce biomechanical effects increase permeability BBB. was able safely temporarily permeabilize acoustic power 5-10 times lower than applied conventional microbubbles, deliver small large molecules into brain.
