摘要: Excised seedling leaf segments of winged bean [Psophocarpus tetragonolobus (L.) DC.] underwent direct somatic embryogenesis under appropriate incubation conditions. Initiation and development the embryos occurred using a two-step culture method. The procedure involved for 28 days on MS basal medium supplemented with 0.1-0.5 mg/l NAA 1.0-2.0 BA (induction medium) before transfer to 0.1 IAA 2.0 (embryo medium). initial exposure low levels coincident high in induction was essential embryogenic induction. Maximum (43.3%) obtained 0.2 BA, at least 14 were required prior embryo medium. conversion frequency cotyledonary 53.3% upon containing ABA 7 followed by IBA BA. Following conversion, regenerated plantlets transferred soil showed normal morphological characteristics.
