摘要: The small GTPase Rab6 is a key regulator in the retrograde transfer from endosomes via Golgi to ER. Three isoforms of have been identified, ubiquitously expressed Rab6A and Rab6A', brain specific Rab6B. Recent studies shown that Rab6A' major isoform regulating this transport. Cytoplasmic dynein main motor protein complex for Dynein consists two heavy chains, intermediate four light chains several called roadblock/LC7 proteins or DYNLRB proteins. In mammalian cells chain DYNLRB1 DYNLRB2. We here show with yeast-two-hybrid, co-immunoprecipitation pull down specifically interacts all three co-localises at Golgi. This first example direct interaction between complex. Pull experiments showed further preferred association GTP-bound interestingly GDP-bound addition was found apparatus where it EYFP-Rab6 isoforms. putative modulator intrinsic activity GTP-binding vitro we were not able reproduce effect on activity.
