C5a-induced chemiluminescence of human granulocytes and its amplification by a serum factor.

摘要: Purified human C5a elicits a fast chemiluminescence (CL) response from isolated granulocytes in the presence of Lucigenin (bis-N-methylacridinium nitrate). The reaction is inhibitable to more than 90% by superoxide dismutase (SOD) - final concentration 200 micrograms/ml -, about 60% catalase 10 mg/ml and 30% hydroxyl radical scavenger D-mannit 100 mM. Therefore O2- seems be oxygen responsible for most CL, while OH H2O2 are also involved. Addition normal pool serum cells 1-2 min before stimulation with strongly enhances effect dose time-dependent manner. existence "helper activity" amplifying C5a-induced CL postulated. This is, however, no specific C5a, since responses elicited chemotactic peptide f-met-phe or phorbol-myristate-acetate (PMA) enhanced preincubation serum. In contrast, ConA-induced not but decreased. Therefore, though unique represent general "adjuvans" on granulocytes, restricted certain stimuli.
