Chiral separation of Huperzine A using CE – Method validation and application in pharmaceutical formulations

摘要: The aim of this work is the development, validation and application an MEKC method for chiral separation Huperzine A. A important compound that used to treat Alzheimer's disease. However, only (-)-form biologically active behaves as a potential acetylcholinesterase inhibitor. Therefore, from (+)-form greatest importance. Optimal conditions, regarding resolution analysis time, were established by altering several experimental parameters, such temperature, field strength, pH, type concentration BGE selector. major problem had be solved in study was low intensity efficiency peaks. More parameters examined, addition modifiers, optimize further separation, particularly efficiency. Baseline enantioseparation achieved using 50 mM acetate (pH 5.0), supplemented with 0.2% w/v poly(sodium N-undecanoyl-ll-alanyl-valinate) 10% v/v tert-butanol. Finally, optimum conditions applied pharmaceutical formulation, demonstrate ability control purity (-)-Huperzine formulations.
