Titration of cardiolipin by either 10-N-nonyl acridine orange or acridine orange sensitizes the adenine nucleotide carrier to permeability transition

摘要: Under the action of carboxyatractyloside or fatty acids, adenine nucleotide translocase switches its function from carrier to modulator opening a non-specific pore. In addition effect these agents, this modification in activity is, some way, dependent on influence lipid milieu membrane. Cardiolipin among other membrane phospholipids, one that interacts most with translocase. This work shows 10-N-nonyl acridine orange and orange, probes for phospholipid, modify sensitivity carboxyatractyloside, oleate, palmitate induce permeability transition. The results also show stimulate release mitochondrial cytochrome c, increase labeling by eosin 5-maleimide. Based aforementioned it is proposed due conformational change translocase, induced binding probe cardiolipin.
