Molecular cloning of poliovirus cDNA and determination of the complete nucleotide sequence of the viral genome

摘要: The complete 7410 nucleotide sequence of poliovirus type I genome was obtained from cloned cDNA. Double-stranded cDNA synthesized and inserted into the Pst site plasmid pBR322, three clones were derived that together provided DNA copies entire genome. Two contained inserts 2.5 6.5 kilobases represented all but 5' 115 bases RNA. A third clone generated primer-extended sequences end viral An open reading frame identified in starting 743 RNA extending to a termination codon 71 3' known polypeptide sequence.
