Towards a metabolic therapy of cancer

摘要: Abstract. It is increasingly appreciated that cancer cells must be endowed with specific metabolic adaptations to support enhanced growth and ensure survival under stressful conditions. On the other hand, many oncogenic mutations of protooncogenes tumor suppressor genes directly cause derangements and, conversely,mutations enzymes have been found underlie several forms cancer. Thus, cancer-specific alterations are now considered among hallmarks malignant tumors. Most commonly, exhibit glycolysis aerobic conditions (the Warburg effect) but in metabolism amino acids, such as glutamine, serine proline described important features selected types. In theory, all these deranged pathways may constitute novel therapeutic targets, although only “metabolic” drug clinical use still represented by enzyme L-asparaginase. However, increasing amount experimental evidence, well number trials progress, suggests drugs will soon complement standard anti-cancer chemotherapy modern biological drugs.
