摘要: Analytical ultracentrifugation (AUC) is one of the classic methods in study biological macromolecules. Developed by Svedberg 1920s, it was a central tool to show, for first time, existence macromolecules solution (Svedberg and Fahraeus, 1926). Owing commercial instrument developed Pickels (Elzen, 1988), widespread use following decades physicochemical properties proteins nucleic acids, including their size, shape, reversible complex formation (Schachman, 1959). Although 1970s 1980s AUC mainstream biochemistry laboratories went into temporary decline, preparative centrifuges had been spawned continued represent basic most biochemical 1988). In 1990s, with increasing availability sophistication recombinant protein expression, general interest interactions emerging, improved instrumentation modern computational capabilities making sedimentation interacting systems more accessible, experienced renaissance, as recognized that ultracentrifuge could provide important unique information on 1989, 1992). this book, two chapters are devoted describe biophysical systems; velocity (SV) AUC, which examines time course sedimentation, equilibrium (SE) (see Chapter 10), concerned thermodynamic analysis final macromolecular distribution.
