Determination of HER-2 status on FNAC material from breast carcinomas using in situ hybridization with dual chromogen visualization with silver enhancement (dual SISH).

摘要: During the last years, HER-2 status kits and protocols for chromogen visualization of hybridization signals have come on market. The first generation using used single color probes. second generation, now emerging market, uses dual visualization. aim this study has been to test a new kit (Ventana INFORM HER2 Dual Colour ISH Roche®) compare results with our in-house method(s). material consisted primarily cytological from invasive breast carcinomas in 49 women. SISH was done all histological specimens. specimens were treated according manufacturer’s recommendations. procedure modified several steps order adapt it material. Hybridization failed two showed concordant as amplified/not amplified. included cases had same expression situ components histology. Four IDC amplification (8.5%). Polysomy found cases. All except one concurred method(s) (1/47=2.1%). is suitable examination status. protocol must be optimized
