摘要: Apolipoprotein (apo) B-100 and B-48 are prominent apolipoproteins in VLDL, IDL, chylomicrons. Organ cultures of normal adult human liver were established to ascertain the form apo B synthesized by hepatocytes humans. Human was minced incubated 15 mL methionine-free RPMI-1640 medium with 10% dialyzed fetal calf serum plus 250 microCi 35S-methionine for eight hours at 37 degrees C. Lipoproteins secreted isolated ultracentrifugation content newly determined quantitation radioactivity apoB-100 apoB-48 bands after separation 3% NaDodSO4 gel electrophoresis. In eight-hour period, 2.5% 3.2% added TCA-precipitable protein which 0.34% B. Ninety-nine percent LDL electrophoretic band. No significant detected Eighty-nine total VLDL 8% IDL LDL, respectively. These results establish that organ culture synthesizes but not B-48.
