摘要: The platelet membrane glycoprotein (Gp) Ib complex consists of four polypeptides: the disulfide-linked GpIb alpha and beta subunits; GpIX, tightly, but noncovalently associated with alpha-beta; more weakly GpV. It is not certain whether association GpIX to Gplb alpha-beta via GpIB alpha, beta, or both subunits, although recently published evidence implicates an interaction beta. We have investigated using polyclonal rabbit antibodies raised by immunization purified glycocalicin synthetic peptide sequences from respectively, as well monoclonal directed against (FMC-25) (AP-1). performed two types experiments, either platelets. (1) When wells were coated nonreduced complex, binding FMC-25 was inhibited 73% antibody, only 30% antibody; when reduced same 86% 13%, respectively. then incubated captured reactivity. reacted strongly; in contrast, reactivity retained complex.
