摘要: Abstract An enzyme-linked immunosorbent assay (ELISA) has been developed for the detection of circulating anti-neutrophil cytoplasm antibodies (ANCA), which are defined by a diffuse, granular staining alcohol-fixed human neutrophils indirect immunofluorescence (IIF). Detection high sensitivity and specificity active Wegener's granulomatosis (WG) reflects effect treatment. In present assay, immunoplates were coated with cytoplasmic α fraction obtained from apparently healthy donors nitrogen bomb cavitation subsequent Percoll gradient centrifugation. Alkaline phosphatase-labelled anti-human IgG was used as secondary antibody. Diluted sera 70 patients WG 16 other diseases anti-myeloperoxidase (anti-MPO) examined. It is concluded that ELISA accurately detects IIF ANCA positive WG, helpful in detecting remission, not influenced presence anti-MPO may help cases granulocyte-specific anti-nuclear since this pattern obscures patterns. The titration can be carried out 3.5 h whereas rapid test just to detect performed 30 min.
