摘要: The mRen2.Lewis congenic strain is an estrogen-sensitive model of hypertension whereby estrogen depletion produces a significant and sustained increase in blood pressure. recent identification G protein-coupled receptor 30 (GPR30) as third isotype prompted us to test the hypothesis that this novel exhibits beneficial cardiovascular actions hypertensive female rat. Intact female, ovariectomized (OVX), male rats were treated with selective GPR30 agonist G-1 or vehicle via osmotic minipump for 2 wk. significantly reduced systolic pressure OVX (178 +/- 7 142 10 mm Hg, P 0.05, n = 5) (207 192 5 > 7). did not alter uterine body weight OVX, suggesting activation distinct from receptor-alpha -beta. In isolated aortic rings constriction response angiotensin II. Vascular angiotensin-converting enzyme type 1 mRNA also lower, whereas enzyme-2 was increased. treatment associated alterations either endothelial nitric oxide synthase expression acetylcholine-induced relaxation. Immunohistochemical staining evident both intima media aorta. We conclude may contribute through regulation vascular components renin-angiotensin system.
