Southern blot patterns, frequencies, and junctional diversity of T-cell receptor-delta gene rearrangements in acute lymphoblastic leukemia

摘要: Southern blot analysis of T-cell receptor (TCR)-delta gene rearrangements is useful for diagnostic studies on the clonality lymphoproliferative diseases. We have developed 18 new TCR-delta probes by use polymerase chain reaction (PCR) techniques. Application these detailed genes in normal control samples, 138 acute lymphoblastic leukemias (T-ALL), and 91 precursor B-ALL allowed us to determine restriction map five enzymes, as well enzyme patterns all theoretically possible rearrangements. Based this information, it appeared that 97% 213 detected our series ALL could be TCRDJ1 probe majority (76%) identified precisely. In T-ALL, we found a strong preference complete V delta 1-J 1 (33%), 2-J (10%), 3-J (7%) incomplete rearrangement D 2- J (11%). B-ALL, consisted 2-D 3 (72%) (10%). The junctional diversity 6 preferential was analyzed showed an extensive insertion (approximately 30 nucleotides) delta-J rearrangements, whereas had correspondingly smaller regions. presented here, combination with are important ALL; can Identification prerequisite subsequent PCR TCR- regions, eg, detection minimal residual disease during follow-up patients.
